I wonder... depending on how vampires work in the COTCverse, the total-replacement transfusion might work, if the transformation isn't complete yet. Might.

There will probably be long-term side effects, though. If only because Jamie is a card-carrying member of the GoEC and needs to torture his characters just that much more.

Well, in one vampire movie (Lost Boys I think) the transformation isn't complete until you drink blood. Until that point you're a half-vampire and can be transformed back to human if the Master Vampire is slain.

Of course, if this were the case, then you'd need to kill Dracula himself in order to free Diane. As Dracula is pretty much unkillable (he can be ashed, but will return with the proper ceremony and his ashes) and there's no sign of the True Cross to be seen... well, Diane is in vamp territory for the unforeseen future.

Which of course will bring about a spin-off comic for some fan to draw... My Mother Is A Vampire. ;)

Unless of course Jamie kills her off permanently. After all, every major death we've had has come back except for Chelsea, and even she's on her way back. :D And Jamie did say someone was going to die... (did he say it would be permanent though? *ponders*)
