Pretty simple, was Justin's "Maybe guys aren't for you?" comment a prophetic, prescient hint at the future, or a joke that became something more?

Considereing Dan once wanted Ellen to be a villain and that Ellen's the reason for Nanase realizing it... Well, there is a slight possibility that it 'grew' outside the Masterplan, but I think Dan had something else Planned for Nanases epiphany (sp?) if it hadn't been for ellen.
Thus yes, it was in the Masterplan.

I think 'Yes' because Dan, when he first started the comic, had "Sister" and "Painted Black" on his radar. "Painted Black" came some time into the comic, and I think by the time "Sister" happened, he had the interim sketched enough to know what was going to happen during "Night Out" and foreshadow some in the beginning of "Sister". The two storylines are almost next to each other, anyway.

I don't think Justin has any special abilities. I think he was just able to read Nanase better than she was willing to read herself.
As for Dan, yes, I think this was obviously planned from the beginning. Although, considering how much Ellen's potential direction shifted, it's remarkable how convenient that works out for Nanase. (i.e. instead of an evil villain, she gets a bisexual female version of the guy she felt was an ideal boyfriend).

this will end up becoming some thing like the Da Vinci Code or Harry Potter, where by the end of it all, all the loose ends will be tied up. (Dan is just taking his time about it and is also working out the perfect way to end it)
so this will be another great storyline just as some of the books that we have seen that are designed to grow up with the audience. This is my view on the matter anyway.

Considering that Nanase immediately responded with an uncharacteristically vehement denial of any possibility she might be gay, it's fairly obvious that her preference was planned out (at least somewhat) by that point.

The "vehement denial = tacit confirmation" is a bit cliched, unfortunately. The only other thing that comes close to being that typical is the running joke that everyone thinks Character X is gay even though he's not; we see some of that too, with Tedd. Nanase's reaction is a bit like a reverse-Melissa, so it's not totally boring.

I'm not sure whether the question was "Is Justin psychic?" or "Was Justin's joke foreshadowing or coincidental?". I've already answered the latter; in the case of the former, he's just a perceptive sort of guy.
